تعیین دوز مؤثر بیهوشی دو ماده MS-222 و 2- فنوکسی اتانول و مقایسه اثر آن‌ها بر برخی شاخص‌های بیوشیمیایی خون ماهی شیربت (Barbus grypus)

چکیده

با توجه به عوارض اجتناب ناپذیر داروهای بیهوش کننده در آبزی پروری، بررسی های خونی و بیوشیمیایی یکی از راه های بسیار مناسب برای شناسایی اثرات جانبی آنها است. هدف از این مطالعه مقایسه اثر غلظت و زمان دو ماده بیهوشی 2- فنوکسی اتانول و MS-222 بر شاخص‌های استرس )گلوکز و کورتیزول) خون ماهی شیربت (Barbus grypus) بود. برای این منظور تعداد 100 قطعه ماهی شیربت با میانگین وزن 5±50 گرم برای بیهوشی با ماده 2-فنوکسی اتانول با غلظت‌های 200، 300 و 400 میکرو لیتر در لیتر و  برای ماده بیهوشی MS222 با غلظت‌های 40، 50 و 80 میلی‌گرم در لیتر و یک گروه نیز به‌عنوان شاهد در نظر گرفته شد. برای هر ماده پس از رسیدن ماهیان به مرحله 4 و 5 بیهوشی، زمان بیهوشی ثبت و سپس در غلظت مؤثر خون‌گیری در فواصل زمانی 3 دقیقه، 6 ساعت، 12 ساعت و 24 ساعت انجام شد. با توجه به نتایج به‌دست‌آمده دوز مؤثر بیهوشی برای 2-فنوکسی اتانول 300 میکرو لیتر/لیتر و برایMS222  50 میلی‌گرم/لیتر محاسبه شد. همچنین نشان داده شد ماده بیهوشی MS222 و 2-فنوکسی اتانول از دقایق اولیه پس از بیهوشی ماهی با افزایش هورمون کورتیزول سبب افزایش میزان گلوکز خون می‌شوند این اثرات افزایشی تا 24 ساعت پس از بیهوشی نیز در خون ماهی دیده می‌شود. مقایسه دو ماده بیهوشی بر فاکتورهای استرس نشان داد تأثیر هر دو ماده سبب افزایش فاکتورهای استرس شده اما اثرات افزایشی 2-فنوکسی اتانول بیشتر است و بین کورتیزول و گلوکز همبستگی مستقیمی وجود دارد. لذا دو ماده بیهوشی بکار رفته سبب القاء استرس در گونه موردمطالعه شده است.
