اثرات ژرانیول و ورزش بر حافظه و یادگیری در موش های نر صحرایی آلزایمری شده

چکیده

بیماری آلزایمر از مهمترین بیماری­های تحلیل برنده عصبی است که به دنبال تولید رادیکال­های آزاد و فاکتور‌های التهابی بروز می­کند. ژرانیول که یک مونوترپن طبیعی است دارای اثرات آنتی­اکسیدانی و ضدالتهابی می­باشد. هدف از این مطالعه، بررسی اثرات ژرانیول و ورزش بر حافظه و یادگیری، میزان فعالیت SOD و سطح MDA، میزان نوروژنز و نیز تعداد پلاک­های آمیلوئیدی در موش­های آلزایمری شده نژاد ویستار می­باشد. برای ایجاد مدل آلزایمری، از تزریق دو طرفه  Aβ 1-42درون هیپوکامپ رت­های نر استفاده شد. گروه­بندی شامل: کنترل منفی، کنترل مثبت، ورزش، آلزایمری، آلزایمری + ورزش، آلزایمری + ژرانیول، آلزایمری + ژرانیول + ورزش می­باشد. نتایج نشان داد که میزان فعالیت SOD و سطح سرمی MDA در گروه­های ورزش، درمانی و دریافت کننده ژرانیول توام با ورزش در مقایسه با سایر گروه­ها به ترتیب افزایش و کاهش معناداری پیدا کردند (001/0>P). نتایج تست رفتاری شاتل باکس نیز نشان داد در مدت زمان ورود به محفظه تاریک، بین گروه­های درمانی و دریافت کننده ژرانیول توام با ورزش در مقایسه با گروه­های آلزایمری و آلزایمری توام با ورزش در سطح (001/0>P) افزایش معناداری وجود داشت. همچنین میزان نوروژنز درگروه­های درمانی و ژرانیول توام با ورزش در مقایسه با گروه­های آلزایمری و آلزایمری توام با ورزش در سطح (001/0>P) افزایش معناداری یافته بود. در تعداد پلاک­های آمیلوئیدی درگروه­های درمانی و ژرانیول توام با ورزش در مقایسه با سایر گروه­ها نیز در سطح (001/0>P) کاهش معناداری مشاهده گردید. با توجه به نتایج به دست آمده در این تحقیق به نظر می­رسد استفاده توام ژرانیول و ورزش می­توانند اختلالات ایجاد شده در حافظه،  میزان فعالیت SOD، سطح MDA، میزان نوروژنز و نیز پلاک­های آمیلوئیدی ایجاد شده در بیماری آلزایمر را به طور چشمگیری بهبود بخشد.
