بررسی اثر سینامالدئید بر تغییرات بیان ژن هایBax ، Bcl2 وTGF-B در سلول های سرطانی شده بافت معده موش توسط دی متیل هیدرازین

چکیده

مطالعات نشان می­دهند Bax و Bcl2 به عنوان ژن­های بسیار مهم در فرآیند آپوپتوز می­توانند از بروز سرطان جلوگیری کنند. نقش ژن TGF-β نیز به عنوان یک ژن  مهم در فرآیند متاستاز در سرطان­ها به اثبات رسیده است. هدف از این بررسی ارزیابی تغییرات بیانی این سه ژن در سلول­های سرطانی شده بافت معده موش توسط دی متیل هیدرازین می­باشد. در این مطالعه تعداد 40 سر موش نژاد ویستار نر 53 هفته­ای بالغ مورد ارزیابی قرار گرفتند و در چهار گروه 10 تایی تقسیم شدند. گروه شاهد فقط آب و غذا دریافت کردند .گروه دوم کارسینوژن بدون سینامالدهید، گروه سوم فقط سینامالدهید و گروه چهارم کارسینوژن همراه با سینامالدهید را همزمان دریافت کردند. سپس بافت معده  جداسازی شده و بیان سه ژن باReal time PCR  ارزیابی شد. داده­ها با نرم افزار SPSS و آزمون آماری one-way ANOVA  مورد بررسی قرار گرفت. بررسی نتایج  نشان داد که بیان ژن­های مذکور در دریافت کننده کارسینوژن همراه با سینامالدهید نسبت به گروه شاهد برایBax  افزایش و برای  Bcl2کاهش معناداری داشت اما برای  ژن TGF-β معنادار نبود. نتایج پژوهش نشان داد که سینامالدهید می­تواند با تغییراتی که در بیان ژن­های Bax، Bcl2 و TGF-β  در بیماری سرطان معده القاء شده توسط دی متیل هیدرازین داشته باشد، پس می­توان امید داشت در تحقیقات تکمیلی سینامالدهید به عنوان یک کاندید در درمان طب مکمل مد نظر قرار گیرد.
