چکیده
سرطان روده بزرگ یکی از شایع­ترین تومورها در جمعیت انسانی است. داروهای زیادی جهت درمان انواع مختلف سرطان در بازار موجود می­باشد ولی به علت اثرات سمی و تاثیرات جانبی استفاده از آنها محدود شده است و در دهه اخیر متخصصین این حوزه بدنبال جایگزین مناسب برای داروهای ضد سرطانی رایج می باشند. هدف از این مطالعه ارزیابی مقایسه اثرات داروی اگزالی پلاتین بر میزان نیتریک اکساید در رده سلولی سرطانی کولورکتال SW480 و غیر سرطانی HEK293 انجام شده است. در این مطالعه تجربی، اگزالی پلاتین در غلظت­های 95/1، 9/3، 8/7، 6/15، 25/31، 5/62، 125، 250 و 500 میکروگرم در میلی­لیتر تهیه شد و اثر سیتوتوکسیک بر روی سلول­های SW480 و HEK293 با استفاده از روش MTT بررسی شد. اکسید نیتریک با استفاده از روشGriess  اندازه­گیری شد و از روش Real Time PCR برای ارزیابی بیان ژن iNOS استفاده شد. در نهایت، از آزمون آنالیز واریانس یک طرفه و آزمون t برای تجزیه و تحلیل داده­ها استفاده شد. نتایج این مطالعه نشان داد که اگزالی پلاتین در غلظت­های 8/7 تا 250 میکروگرم بر میلی لیتر روی سلول­های  SW480 اثر کشنده­ای بر روی سلول دارد اما دربقیه غلظت­ها هیچ اثر سمیت سلولی بر روی رده­ی سلولی طبیعی نشان نداد. اگزالی پلاتین در غلظت­های 10، 20 و 40 میکروگرم در میلی­لیتر تولید اکسید نیتریک در رده­های سلولی  SW480به طور قابل توجهی متفاوت از سلول­های  HEK293به عنوان رده سلولی نرمال است. نتایج Real Time PCR نشان داد که بیان ژن iNOS در سلول­های SW480 و HEK293 تفاوت معنی داری با گروه کنترل ندارد (05/0 p >).
