بررسی تاثیر دیازینون بر تغییرات هیستومورفومتریک هیپوکامپ جنین موش کوچک آزمایشگاهی نژاد Balb/c

چکیده

دیازینون دسته‌ای از ارگانوفسفره­های آفت­کش است که بطور وسیع در کشاورزی استفاده می شود. این ماده سمی از طریق تنفسی، پوستی و گوارشی جذب می­شود و بر بافت های مختلف تاثیر می­گذارد. بنابراین هدف از این تحقیق بررسی اثرات دیازینون بر هیستومورفومتری هیپوکامپ جنین موش کوچک آزمایشگاهی نژاد Balb/c می­باشد. تعداد 25 سر موش کوچک آزمایشگاهی ماده بالغ نژاد c/Balb به 5 گروه 5 تایی تقسیم شدند.گروه شاهد دیازینونی دریافت نکرد. گروه­های شم الف و ب، امولسی فایر را به ترتیب به مقدار 52/0 و 2/5 میکرولیتر در واحد حجم (5000 سانتی متر مکعب در دسیکاتور) و گروه‌های تجربی الف و ب، دیازینون را به ترتیب به مقدار 3/1 و 13 میکرولیتر در واحد حجم، از روز 7 تا 18 بارداری، یک روز در میان بصورت استنشاقی دریافت کردند. در پایان روز 18 بارداری موش­ها کشته شده، جنین­ها از بدن حیوان خارج گردیده، از نظر ظاهری بررسی شدند. پس از فیکس شدن و پردازش جنین­ها، مقاطع 5 میکرونی تحت رنگ­آمیزی هماتوکسیلین-ائوزین قرار گرفت و بررسی هیستومورفومتری از هیپوکامپ انجام شد. میانگین ضخامت لایه‌‌های مختلف CA1، CA2 و CA3  هیپوکامپ در گروه‌های تجربی الف و ب کاهش داشت. شمارش میانگین تعداد سلول­ها در لایه‌‌های مختلف هیپوکامپ و شکنج دندانه‌ای در گروه تجربی الف و ب افزایش نشان داد. به نظر می­رسد که استنشاق دیازینون در مقادیر بالا (دوز 13 میکرولیتر در واحد حجم) در موش باردار می­تواند سبب تغییرات هیستومورفومتریک در روند تکامل هیپوکامپ گردد.
