تعیین اثرات هروئین بر روی ترکیبات کیسه آمنیوتیک در موش های باردار و تخمدان فرزندان آن‌ها

چکیده

هروئین یکی از اعتیاد­آورترین مواد مخدری است که در حال حاضر در ایران در میان نوجوانان و جوانان مورد سوء مصرف  قرار می‌گیرد. از آن‌جا که استفاده از مواد مخدر می‌تواند اثرات زیان­باری بر روی اعضاء بدن، بافت‌ها، هورمون‌ها، مایع آمنیوتیک، بیان ژن های مؤثر در مادر و جنین، و نیز سایر فاکتورها داشته باشد، بنابراین بررسی اثرات هروئین بر کیسه آمنیون موش‌های باردار و تأثیرات آن بر نسل‌های بعد آن نیز بسیار ضروری می‌باشد. 60 سر موش صحرایی بالغ ماده نژاد ویستار به 6 گروه کنترل، شم، دریافت کننده دوز 1 میلی­گرم/کیلوگرم هروئین، دریافت کننده دوز 2 میلی­گرم/کیلوگرم هروئین، دریافت کننده دوز 3 میلی­گرم/کیلوگرم هروئین و دریافت کننده دوز 4 میلی­گرم/کیلوگرم هروئین تقسیم شدند. در انتها، خونگیری از قلب موش‌ها انجام شد و پارامترهای تستوسترون، گلوکز، LH،  FSH، AST، ALT،  WBCوRBC  اندازه گیری شدند. در ادامه، مایع آمنیوتیک جهت سنجش میزان کربوهیدرات­ها، فسفولیپید، چربی­ها، پروتئین و اوره جدا شد. در پایان، داده‌ها از نظر آماری مورد ارزیابی قرار گرفتند. تزریق درون صفاقی هروئین تغییرات قابل توجهی را در میزان پرامترهای تستوسترون و تری‌گلیسیرید نسبت به گروه کنترل ایجاد نمود. این در حالی است که در میزان LH و FSH تغیر قابل توجهی مشاهده نشد. همچنین افزایش بیان ژن‌های در نتیجه افزایش دوز هروئین دیده شد. در این بررسی تزریق هروئین حتی در دوزهای پایین توانست باعث افزایش یا کاهش فعالیت تخمدان و اووژنز و در نتیجه اختلال در بلوغ اووسیت‌ها و کیفیت تخمک‌گذاری گردد.
